    As the Senior Associate Scientist, you will join efforts focused on the research of novel agents to treat help patients' own immune systems fight their cancer. You will interact with external and internal colleagues as a key member of a multidisciplinary team to support drug discovery and development. You will also be responsible for designing and implementing novel assays to evaluate performance, mechanism of action, and resistance characteristics of novel agents, and will assist in the development of new tools to study and treat disease.

    KEY RESPOSIBILITIES:

    1. Support the identification, validation, and drug development of novel immuno-oncology therapeutics for the treatment of Oncology indications
    2. Design, execute and interpret studies for the identification, validation, and characterization of antigen-specific T cell immune responses
    3. Establish immunogenicity assays with proliferation and cytokine endpoint readouts for the characterization of immune responses to both neo and self-antigens
    4. Design, execute, and interpret complex in vitro, ex vivo, and in vivo experiments to evaluate mechanism of action associated with immune cell function with the aim of advancing the Oncology pipeline.
    5. Communicate scientific results to multidisciplinary teams and project leader. Comfortable working in a highly matrixed environment
    6. Maintain detailed and organized experimental records and stay compliant on all relevant internal and external training requirements

    OTHER RESPONSIBILITIES / DETAILED DUTIES:

    1. Quantify immune cell response by Elispot/fluoropspot/MSD/ELISA (e.g. IFNγ, TNFα, IL-2, IL-10)
    2. Analyze, compare, and report the sensitivity, potency, and impact on growth rate of molecules of interest agents by standard cytotoxicity assays (e.g. Incucyte, Cell titer-glo, LDH, MTT)
    3. Quantitate soluble cytokine and protein levels from serum, tissue homogenate, and tissue culture supernatant by standard ELISA and other platforms (e.g. MSD, Luminex) including utilization of Spotfire analysis software
    4. Culture multiple primary cell types, and cancer cell line simultaneously under aseptic conditions
    5. Establish robust assay readout metrics (EC50/IC50, max response, response above background) for the comparison and ranking of experimental agents
    6. Process human or mouse tissue (blood, lymph nodes, tumor, prostate, lung, muscle, spleen, and thymus) to single cell suspension for analysis of immune cell and stromal cells by Flow cytometry
    7. Stain and characterize immune cell population by multiparameter (8-16 colors) flow cytometry (e.g. Fortessa, Canto, Celesta)

  • Qualifications
    • A Master’s degree or equivalent in the Life Sciences (molecular/cellular biology, immunology, pharmacology) with 3 or more years of experience, or a Bachelor’s degree with 5 or more years of experience is required.
    • Experience in design and interpretation of cell based assays/experiments to interrogate questions in Immunology and compound effects on the immune system is required.
    • Experience culturing multiple primary and cancer cell lines simultaneously under aseptic conditions is required.
    • Experience with cytokine expression, proliferation, and cytotoxicity assays is required
    • Experience in molecular biology including RNA, DNA and protein analysis is required.
    • Experience with multiparameter flow cytometry (8-16 color; Fortessa, Canto, Celesta) of human and murine immune cell populations and analysis using FlowJo software is required.
    • Detail-oriented, with excellent record-keeping skills is required.
    • Experience utilizing Elispot/Fluorospot/MSD/ELISA to quantify immune cell activation including Spotfire analysis software is preferred.
    • Familiarity with gene transduction systems (lenti, adeno, CRISPR-Cas9/12, etc) is preferred.
    • Excellent communication and collaborative skills are essential including; interfacing with group leaders to ensure that in vitro and in vivo testing requirements and timelines are met; developing and presenting summaries of data, writing clear and succinct study reports; and effective interfacing with other cross functional groups.
    • Travel is up to 10%
