    Post-Doctoral Fellow for Immuno-Therapy Pathway Area Stronghold (IT PAS) group within Oncology Discovery at Janssen Research & Development will lead mechanism of action (MOA) studies to enhance our understanding of therapeutic index of immunotherapy assets alone and in combination with SOC and agents in areas of interest to our oncology portfolio.  The length of this postdoctoral fellow opportunity will be 24-36 months.

     

    In this role you will:

    • Lead and support studies of key Mechanism of action (MOA), mechanisms of resistance and best combinatorial strategies to guide development of immuno-oncology therapeutics for the treatment of Oncology indications
    • Support the identification, development, and implementation of biomarker strategies for oncology immuno-modulatory compounds in stages from discovery to early and mid- clinical development
    • Design, execute and interpret complex in vitro, ex vivo and in vivo experiments to evaluate mechanism of action associated with immune cell function
    • Communicate scientific results to multidisciplinary teams and project leader. Comfortable working in a highly matrixed environment
    • Culture multiple primary cell types, and cancer cell line simultaneously under aseptic conditions
    • Establish robust assay readout metrics (e.g. EC50/IC50, max response, response above background) for the comparison and ranking of experimental agents
    • Process tissue (blood, lymph nodes, tumor, prostate, lung, muscle, spleen, and thymus) to single cell suspension for analysis of immune cell and stromal cells by Flow cytometry or spectral cytometry
    • Stain and characterize immune cell population by multiparameter (8-16 colors) flow cytometry (e.g. Fortessa, Canto, Celesta)
    • Maintain detailed and organized experimental records and stay compliant on all relevant internal and external training requirements
    • Collaboration in various departments across Janssen including oncology, immunology and discovery, and translational research
    • Can collaborate across prominent academic labs and recognized experts studying immune modulation, genetic alterations, and novel technologies in cancer.
    • Perform bioinformatics and statistical analysis, alternative visualization methods, and data interpretation of public and proprietary data for the Immuno-Therapy Pathway Area Stronghold (IT PAS).
    • Integrate data from external (e.g. TCGA, NCBI Entrez, NCBI GEO, UCSC Genome Browser) and internal sources in analyses.
    • Work independently as part of a matrix organization.
    • Communicate results to computational biologists, bench scientists, clinicians, and management in an appropriate biological context.
  • Qualifications
    • A PhD degree in Cell Biology, Immunology, or tumor immunology field received no more than 3 years ago
    • A demonstrable record of scientific publications is required if coming from an academia. 
    • A strong background in characterization of immune cell population by multiparameter (8-16 colors) flow cytometry (e.g., Fortessa, Canto, Celesta), CYTOF, or spectral cytometry is required
    • Expertise and deep understanding of the complexity of the tumor micro-environment including major roles for Tregs, TAM/TAN, CAFs, MDSCs, T cell, NK, gd, and immuno-modulatory pathways active in the TME is highly preferred.
    • Expertise in design and interpretation of T-cell antigen specific immunogenicity assays/experiments is required (e.g., Elispot, ICS, flow cytometry)
    • A proven track record applying a variety of Cellular, and Immunology based techniques to isolate, phenotype and functionally test peripheral and tumor infiltrating lymphocytes is required.
    • Expertise in molecular biology including RNA, DNA and protein analysis is preferred.
    • Experience with in vivo models of tumor biology is highly preferred.
    • Detail-oriented, with excellent record-keeping skills is required.
    • Strong written and verbal communications skills and collaborative skills are essential.  Including interfacing with group leaders to ensure that in vitro and in vivo testing requirements and timelines are met; developing and presenting summaries of data, writing clear and succinct study reports; and effective interfacing with other cross functional groups.
    • Must be legally authorized to work in the United States and not require future sponsorship for employment visa status now or in the future.
    • This role is to be located in Spring House, PA and may require up to 10% travel domestic or internationally.
