ADENO-ASSOCIATED VIRUS PREFORMULATION INTERNSHIP OVERVIEW

Recombinant adeno-associated virus (AAV) vector has been the most popular vector choice for in vivo gene therapy, due to its long-term expression and lack of pathogenicity in humans. Hundreds of ongoing clinical trials, utilizing different AAV serotypes, target multiple genetic diseases such as cystic fibrosis and hemophilia and eye diseases such as age-related macular degeneration (AMD). At AbbVie, rAAV is a growing therapeutic modality, requiring an increase of in-house development knowledge. However, the developability assessment and preformulation development of AAV are challenging due to limited material availability and AAV structure complexity. Both the capsid and its packaged DNA cargo are potential sources of AAV instability. There is an urgent need for detailed characterization of the physicochemical stability of AAVs to enable rapid screening and early drug product development.
KEY RESPONSIBILITIES INCLUDE:
Develop stability-indicating AAV characterization methods that are suitable for the small amounts of materials typically available at late-discovery/early-development stages
Develop a representative platform formulation for physical and chemical stability evaluation of AAV candidates under stressed conditions and in-use condition
MINIMUM QUALIFICATIONS
Currently enrolled in university, pursuing a Bachelor’s, Master’s, or PhD in Biology, Chemistry, Biophysics, Biochemistry, Biopharmaceutics, Pharmaceutical Sciences, Chemical Engineering or related field
Must be enrolled in university during the internship period
PREFERRED QUALIFICATIONS
Currently enrolled in PhD program
Exposure to protein biophysics
Laboratory experience in protein formulation
