    BIOLOGICS DEVELOPMENT INTERN NEW BRUNSWICK, NJ

    PROJECT DESCRIPTION: In stem cell biology, differentiation and dedifferentiation are governed by transcriptional changes as well as by epigenetic changes. Recent publications show that certain epigenetic changes during single cell cloning in CHO cells are correlated with improved titers in clones, suggesting that these changes are inherited during cell division, and therefore stable. The dedifferentiation state, such as it happens in the induced pluripotent stem cells, can be achieved and maintained by small molecules supplemented in medium. Here, we propose to compile a comprehensive literature search to identify factors and/or small molecules that lead to epigenetic changes. Identified candidate molecules can then be supplemented into cloning medium during single cell cloning of transfected CHO cells. The aim will be to test the impact of these factors and small molecules in enhancing the specific productivity of monoclonal antibodies in CHO production cells. Business Justification for the Project Enhancing the specific productivity of production CHO cell lines. Key Deliverables at Project Completion

    • A comprehensive list of small molecules and factors that can re-program cells through epigenetic changes.
    • proposal of a single cell cloning experiment with supplementation of the most anticipated factors.

    • Provide additional details about how you will engage and support the student to build their interest in both Biopharma careers and the Client as pre-eminent employer in this field.
    • This project will introduce the intern to a variety of technical and scientific areas of cell line development including clone selection.
    • The intern will regularly interact with the mentor and sponsor to learn new techniques and obtain a general overview of pharmaceutical process development.
    • This will continue to foster a good relationship between academic programs and the Client while introducing students to projects that improve business timelines in addition to enhancing scientific innovation.

    REQUIRED SKILLS:

    • Stem Cells, Cell-culture, aseptic technique
    • Major: Chemical Engineering, Cell Biology, Regenerative Cell Biology
    • School Year of the Student (for the term of the proposed position): Senior
    • Will also consider Junior and Sophomore year student.
