PROJECT DESCRIPTION: In stem cell biology, differentiation and dedifferentiation are governed by transcriptional changes as well as by epigenetic changes. Recent publications show that certain epigenetic changes during single cell cloning in CHO cells are correlated with improved titers in clones, suggesting that these changes are inherited during cell division, and therefore stable. The dedifferentiation state, such as it happens in the induced pluripotent stem cells, can be achieved and maintained by small molecules supplemented in medium. Here, we propose to compile a comprehensive literature search to identify factors and/or small molecules that lead to epigenetic changes. Identified candidate molecules can then be supplemented into cloning medium during single cell cloning of transfected CHO cells. The aim will be to test the impact of these factors and small molecules in enhancing the specific productivity of monoclonal antibodies in CHO production cells.
