Senior / Principal Scientist, Protein Analytics (Biophysical Characterization & Analytical Methods)

    Position Summary

We are a stealth biotechnology company developing precision therapeutics for cardiovascular disease, seeking a highly skilled and hands-on Senior to Principal Scientist to join our Protein Science team.

This role will focus on analytical and biophysical characterization of complex protein–oligonucleotide conjugates (AOCs). The ideal candidate will have experience with oligonucleotide payloads (ASO, siRNA, PMO) and associated analytical workflows, including mass spectrometry, chromatography, and stability assessment. The successful candidate will be a core technical contributor, working closely with colleagues across Protein Chemistry, Biology, and CMC to generate decision-grade analytical data packages that enable rapid iteration and advance conjugate molecules from early discovery through preclinical development.

This position is ideal for a scientist who thrives at the bench, enjoys developing analytical workflows, and can interpret complex datasets to drive clear recommendations. The role is expected to deliver discovery-stage analytical data packages and recommendations to guide conjugation optimization, purification strategy, and molecule design/selection.

    Key Responsibilities

Analytical Strategy & Data Packages • Develop and apply analytical workflows to assess identity, purity, heterogeneity, aggregation, and stability of protein conjugates. • Deliver analytical data packages that guide conjugation optimization, purification strategy, and molecule design and selection.

Mass Spectrometry & Chromatography • Perform and interpret LC-MS and chromatography-based analyses to assess identity, purity, conjugation outcomes, and heterogeneity (SEC, IEX, LC-MS). • Troubleshoot and refine analytical workflows to improve robustness, throughput, and interpretability. • Maintain and troubleshoot analytical instrumentation as needed, and coordinate routine maintenance/service.

Affinity & Biophysical Characterization • Run and interpret affinity methods (SPR/BLI) to assess binding activity and kinetics. • Perform biophysical characterization to assess developability and aggregation risk (for example, DLS, DSF, SEC-MALS).

Stability & Stress Testing • Design and execute stability and stress-condition assessments to understand liabilities and guide lead selection.

Cross-Functional Collaboration • Work closely with Protein Chemistry, Biology, and CMC teams to align analytical strategy with downstream in vitro and in vivo studies and development needs. • Communicate clearly and proactively on timelines, risks, and data-driven recommendations.

Innovation & Platform Development • Contribute experimental data and ideas toward advancement of the AOC analytics platform and related intellectual property. • Develop and document workflows (SOPs) and support method or process transfer as needed (internal or external).

    Qualifications

Required: • Ph.D. in Analytical Chemistry, Biochemistry, Biophysics, Chemical Biology, or related field. • 3-5 years of relevant experience, level-dependent. • Strong hands-on experience with LC-MS and chromatography-based characterization of proteins/bioconjugates. • Strong hands-on experience with affinity methods (SPR/BLI) and interpretation of kinetic/affinity datasets. • Experience with oligonucleotide chemistry and/or oligonucleotide therapeutics (ASO, siRNA, PMO), including conjugates. • Experience independently designing experiments and driving technical problems to resolution.

Preferred: • Experience supporting production and purification of antibody–oligonucleotide conjugates (mg scale), including chromatography-based purification and polishing workflows, is a plus. • Experience working in early-stage or fast-paced biotech environments.

    Key Competencies

    • Strong experimental rigor and problem-solving skills. • Ability to operate as a senior individual contributor within a multidisciplinary team. • Clear communication of complex technical data to scientific colleagues. • Curiosity, adaptability, and enthusiasm for developing new therapeutic modalities.
